FJK-16s 항체는 mouse, rat, dog, porcine, bovine, cat의 FOXP3 단백질(FORKHEAD BOX P3, SCURFIN, JM2)을 인식합니다. FOXP3는 49–55 kDa 크기의 전사 조절 인자로, CD4+CD25+ 조절 T세포(Treg)에서 높은 수준으로 발현됩니다.
이 항체는 mouse FOXP3 단백질의 75–125번 아미노산 영역을 인식하며, 인간에서는 해당 영역이 mRNA 수준에서 대체 스플라이싱됩니다. FJK-16s는 Foxp3/Transcription Factor Buffer Staining Set (Product #00-5523-00)과 함께 사용하는 것이 권장됩니다.

Flow Cytometry를 통한 세포내 염색에 사용됩니다.
Applications Tested:
Mouse splenocyte의 세포내 염색에 대해 테스트되었으며, 0.5 µg/test 이하로 사용 가능합니다. 최적의 성능을 위해 항체 농도는 실험적으로 조정하는 것이 좋습니다.
Fluorochrome Information:
Alexa Fluor 532는 532 nm Green laser로 여기되어 561 nm에서 방출합니다. Yellow-Green laser(561 nm)에서는 사용 불가하며, 560/14 band pass filter 사용을 권장합니다.
Target Information
FOXP3(Forkhead box protein 3)는 면역 항상성 유지에 필수적인 전사 조절 인자로, CD4+CD25+ 조절 T세포에서 안정적으로 높은 수준으로 발현됩니다.
FOXP3 결함은 마우스에서 scurfy 표현형을 유발하며, 인간에서는 IPEX syndrome(면역기능장애, 다내분비병증, 장병증, X연관 증후군)과 관련이 있습니다.
인간 유방암 및 대장암 세포에서는 DNA 손상에 반응하는 p53에 의해 FOXP3 발현이 조절됩니다.
