Immunohistochemistry (PFA fixed) 시, 항체를 실온에서 30분간 반응시킵니다. 포르말린 고정 조직 염색 시에는 10 mM Tris, 1 mM EDTA, pH 9.0 용액에서 95°C로 45분간 가열 후, 실온에서 20분간 냉각하는 항원 복원 과정이 필요합니다.
Target Information
CD1a는 비다형성(non-polymorphic) MHC Class I 관련 세포 표면 당단백질로, β2-microglobulin과 결합하여 발현됩니다. 주로 피질 흉선세포(cortical thymocytes), 랑게르한스 세포(Langerhans cells), 수지상세포(dendritic cells) 등에서 발현되며, 일부 T세포 계통 악성종양 및 조직구증에서도 관찰됩니다.
CD1 단백질은 자가 또는 미생물 유래의 지질 및 당지질 항원을 T세포에 제시하며, 인간 게놈에는 CD1a, b, c, d, e 등 다섯 가지 CD1 유전자가 존재합니다. CD1a는 주로 세포막과 초기 엔도시토시스 소포 내에서 재순환하며, 대체 스플라이싱 변이체가 보고되어 있으나 그 생물학적 의미는 명확하지 않습니다.
