Target Information
Protein C is a vitamin K-dependent plasma glycoprotein that is cleaved to its activated form by the thrombin-thrombomodulin complex. Activated Protein C contains a serine protease domain and degrades the activated forms of coagulation factors V and VIII. It is produced in the liver as a 62 kDa proenzyme, which is activated by thrombin bound to an endothelial receptor. The active enzyme (55 kDa) cleaves factors Va and VIIIa, inhibiting blood coagulation. Normal plasma concentrations are approximately 1–3 ng/mL for the active enzyme and ~3 µg/mL for the proenzyme. Mutations in the Protein C gene are associated with thrombophilia, neonatal purpura fulminans, and recurrent venous thrombosis.
