Target Information

F-box and WD Repeat Domain Containing 7 (FBXW7) is a member of the F-box protein family, characterized by an approximately 40 amino acid F-box motif. These proteins form part of the SCF (SKP1–Cullin–F-box) ubiquitin ligase complex, which functions in phosphorylation-dependent ubiquitination.
F-box proteins are classified into three groups:

  • Fbws: containing WD-40 domains
  • Fbls: containing leucine-rich repeats
  • Fbxs: containing other interaction modules or no recognizable motifs

FBXW7, also known as FBX30, belongs to the Fbws class and includes seven tandem WD40 repeats. It binds to cyclin E and likely targets it for ubiquitin-mediated degradation. Mutations in this gene have been detected in ovarian and breast cancer cell lines, suggesting a role in cancer pathogenesis. Multiple transcript variants encoding different isoforms have been identified.
