Target Information
MAP4K3 (also known as GLK) is a member of the STE20 family of serine/threonine protein kinases. It contains an N-terminal catalytic domain and a C-terminal regulatory domain. MAP4K3 acts upstream of MEKK1 and is stimulated by UV radiation and TNF-alpha, leading to activation of the JNK signaling pathway. It can directly activate PKC-T during TCR signaling, and enhanced MAP4K3 expression is observed in systemic lupus erythematosus. MAP4K3 functions as a pro-apoptotic kinase that activates BAX via modulation of PUMA, BAD, and BIM. The protein specifically activates the c-Jun N-terminal kinase (JNK) pathway and functions upstream of MEKK1. Previously known as RAB8-interacting protein-like 1 (RAB8IPL1), it has been renamed mitogen-activated protein kinase kinase kinase 3 (MAP4K3).
