Target Information
Epithelial sodium channels (ENaC) are amiloride-sensitive members of the degenerin/epithelial sodium channel (Deg/ENaC) superfamily of ion channels. These channels share structural similarity, possessing two short intracellular termini, two transmembrane segments, and a large extracellular loop with a conserved cysteine-rich region.
There are three homologous isoforms of ENaC: alpha, beta, and gamma. ENaC plays a critical role in sodium and fluid balance across epithelial tissues in the kidney, lung, and colon. It mediates aldosterone-dependent sodium reabsorption in the distal nephron, influencing blood pressure regulation. ENaC activity is also modulated by interaction with the cystic fibrosis transmembrane conductance regulator (CFTR) chloride channel.
Mutations in ENaC subunits are associated with human diseases: gain-of-function mutations in beta or gamma subunits cause Liddle’s syndrome (severe hypertension), while loss-of-function mutations in alpha or beta subunits lead to pseudohypoaldosteronism type I (PHA-1).
