Target Information
The mammalian Phospholipase Cg (PLCg) family includes two closely related proteins, PLCg1 and PLCg2. These isozymes share a core structure and possess a unique array of domains, including an additional PH domain, two SH2 domains, and one SH3 domain.
Upon stimulation by extracellular signals such as hormones and growth factors, receptor tyrosine kinases (RTKs) phosphorylate and activate PLCg. Activated PLCg catalyzes the hydrolysis of phosphatidylinositol 4,5-bisphosphate (PIP2) to generate the second messenger molecules inositol 1,4,5-triphosphate (IP3) and diacylglycerol (DAG).
