Lipids play essential roles in cells, including membrane structure, energy storage, transport of metabolic fuels, cell recognition, and signal transduction.
Many cultured cells can synthesize necessary lipids from acetyl CoA. However, certain myeloma cell lines such as NS0, used for large-scale recombinant protein production, are sterol auxotrophs and lack a key enzyme in cholesterol biosynthesis. These cells therefore require an external sterol source.

In serum-supplemented cultures, serum provides extracellular lipids. In serum-free systems, lipids must be added as supplements or concentrates, which are mixtures of fatty acids, detergents, and lipids essential for cell growth and maintenance.
