Although most cells can synthesize lipids and sterols required for their metabolic processes, supplementation with preformed lipids reduces the need for biosynthesis and increases the efficiency of cell metabolism and growth.

Certain cell lines, such as NS0 (mouse myeloma cell line), require large amounts of cholesterol but are incapable of producing it. These cells therefore need supplementation of culture media with a source of sterol.

Cholesterol supplement is used as an additive in serum-free media.
It serves as an important attachment factor for increased plating efficiency and helps protect cells from shear stress.
