Histone deacetylase 4 (HDAC4)는 핵심 히스톤(H2A, H2B, H3, H4)의 N-말단 라이신 잔기를 탈아세틸화하는 효소입니다. 히스톤 탈아세틸화는 전사 억제의 후성유전적 표지로 작용하며, 전사 조절, 세포 주기 진행, 발달 과정에 중요한 역할을 합니다.
HDAC4는 MEF2A, MEF2C, MEF2D와 같은 myocyte enhancer factor와의 상호작용을 통해 근육 성숙에도 관여합니다. 또한, 유방암에서 ESR1 발현의 MTA1 매개 후성유전 조절에 관여하며, HDAC4 유전자의 돌연변이는 Brachydactyly-mental retardation syndrome (BDMR)과 관련이 있습니다.
