eBioscience의 FOXP3 단클론 항체 패널은 인간/영장류 및 마우스/랫의 다양한 FOXP3 단백질 에피토프를 인식하여 FOXP3 발현 패턴 및 FOXP3+ 세포 하위집단 연구에 유용합니다.
150D/E4 항체는 인간 세포의 Exon 2 스플라이스 영역에 매핑되었으며, 마우스에서는 해당 스플라이싱이 보고되지 않았습니다. 이 항체는 epitope mapping을 통해 특성화되었습니다. PCH101 및 236A/E7 항체는 스플라이스형과 full-length 형태 모두를 인식하며, 150D/E4보다 강한 염색을 보입니다.
150D/E4 항체는 Mouse, Rat, Human의 FOXP3(Forkhead box P3, SCURFIN, JM2)와 반응하며, 다른 단백질과의 교차 반응은 확인되지 않았습니다. FOXP3는 49–55 kDa 크기의 단백질로, scurfy(sf) 마우스에서 결함 유전자로 확인되었습니다. CD4+CD25+ 조절 T세포(Treg)에서 높은 발현을 보이며, CD4+CD25- 세포에서 FOXP3의 발현은 Treg 특성을 유도합니다.

FOXP3(Forkhead box protein 3)는 포크헤드/윙드헬릭스 전사 조절 인자 계열의 단백질로, 면역 항상성 유지에 필수적입니다.
381 아미노산 길이의 FOXP3는 CD25+CD4+ 조절 T세포에서 안정적으로 높은 수준으로 발현되며, CD4+CD25- 세포에서는 낮은 수준, CD4- 또는 CD8+ 세포에서는 발현되지 않습니다.
FOXP3 유전자의 결함은 마우스에서 scurfy 표현형을, 인간에서는 IPEX 증후군(면역기능이상, 다내분비병증, 장병증, X-연관 증후군)을 유발합니다.
또한, 인간 유방암 및 대장암 세포에서 FOXP3의 발현은 p53에 의해 DNA 손상 반응 시 조절됩니다.
