Target Information

Crystallins are divided into two classes: taxon-specific (enzyme) and ubiquitous. The ubiquitous crystallins are the major proteins of the vertebrate eye lens, maintaining its transparency and refractive index. Lens fiber cells lose their nuclei during development, so crystallins are synthesized once and retained throughout life, making them highly stable proteins.

Mammalian lens crystallins are categorized into alpha, beta, and gamma families. Beta and gamma crystallins form a superfamily, with alpha and beta further divided into acidic and basic subgroups. Beta-crystallins are the most heterogeneous, differing by the presence of a C-terminal extension (present in basic group, absent in acidic group). They form aggregates and can self-associate or heterodimerize with other beta-crystallins.

The beta Crystallin A3 gene encodes two proteins (beta A3 and beta A1) from a single mRNA. Beta A1 is 17 amino acids shorter than beta A3 due to an alternative translation initiation site. Deletion of exons 3 and 4 is associated with the autosomal dominant disease "zonular cataract with sutural opacities."
