Target Information
NUAK1 (ARK5) is a serine/threonine kinase involved in multiple cellular functions. Overexpression is associated with tumor invasion and metastasis, particularly in colorectal neoplasms. It regulates cellular senescence and ploidy by phosphorylating LATS1 at Ser-464, influencing its stability. NUAK1 also controls cell adhesion through the myosin protein phosphatase 1 (PP1) complex by phosphorylating PPP1R12A, leading to reduced dephosphorylation of myosin MLC2. It may participate in DNA damage response by phosphorylating p53 at Ser-15 and Ser-392 and promoting transcription activation at the CDKN1A/WAF1 promoter. Additionally, NUAK1 suppresses Fas-induced apoptosis via CASP6 phosphorylation and contributes to UV-induced DNA damage response by phosphorylating CDKN1A with STK11, aiding in its degradation for optimal DNA repair.
