Background
The TCF-1 (Transcription factor 7) protein exists in multiple isoforms (L and S) produced by alternative promoter usage. It functions as a transcriptional activator involved in T-cell lymphocyte differentiation and is essential for CD4(+) CD8(+) immature thymocyte survival. Isoforms lacking the N-terminal CTNNB1 binding domain cannot perform this role.
TCF-1 binds to the T-lymphocyte-specific enhancer element (5'-WWCAAAG-3') in the CD3E promoter and may act as a feedback transcriptional repressor of CTNNB1 and TCF7L2 target genes. TLE1–4 proteins repress transactivation mediated by TCF7 and CTNNB1. Expression is mainly in T-cells, proliferating intestinal epithelial cells, and basal epithelial cells of the mammary gland.
