Background
The STRAD protein contains a STE20-like kinase domain but lacks residues critical for catalytic activity, classifying it as a pseudokinase. It forms a heterotrimeric complex with serine/threonine kinase 11 (STK11/LKB1) and calcium-binding protein 39 (CAB39/MO25), activating STK11 and facilitating its phosphorylation while excluding it from the nucleus. STRAD is essential for STK11-induced G1 cell cycle arrest. Mutations in this gene are associated with polyhydramnios, megalencephaly, and symptomatic epilepsy (PMSE) syndrome. Multiple transcript variants encoding different isoforms have been identified.
