ATP6V1F encodes a component of vacuolar ATPase (V-ATPase), a multisubunit enzyme that mediates acidification of eukaryotic intracellular organelles.
V-ATPase dependent organelle acidification is necessary for processes such as:
- Protein sorting
- Zymogen activation
- Receptor-mediated endocytosis
- Synaptic vesicle proton gradient generation
V-ATPase consists of:
- V1 domain (cytosolic): A₃B₃CDEFG₂H subunits – contains ATP catalytic site
- V0 domain (transmembrane): a, c, c’, c”, and d subunits
Additional isoforms of many subunits are encoded by multiple genes or alternatively spliced variants.
