Target Information
KIT (c-KIT) is a proto-oncogene encoding a type III transmembrane receptor for mast cell growth factor (MGF, also known as stem cell factor). It regulates growth and activation of various hematopoietic and non-hematopoietic cells.
Mutations in KIT are linked to gastrointestinal stromal tumors, mast cell disease, acute myelogenous leukemia, and piebaldism.
Upon ligand binding, c-Kit dimerizes and autophosphorylates tyrosine residues (e.g., Tyr703), enabling docking of Grb2 and activation of the Ras/ERK pathway.
SCF/c-Kit signaling activates multiple downstream pathways including PI3K, PLC-γ, and JAK/STAT, essential for hematopoiesis, stem cell maintenance, and gametogenesis.
