Product Specific Information

Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD86 is one of two ligands (the other CD80) for CTLA4 and CD28. It acts as a costimulatory molecule during antigen presentation, providing T-cell help.
In the absence of sufficient co-stimulation involving CD86/CD80, tolerance may be induced. CD86 is a type I membrane protein belonging to the immunoglobulin superfamily, expressed by antigen-presenting cells.
It binds to CD28 and CTLA4 on T cells:

  • CD86–CD28 interaction provides a costimulatory signal for T-cell activation.
  • CD86–CTLA4 interaction negatively regulates T-cell activation, reducing immune response.

Alternative splicing produces multiple isoforms of CD86. Dysfunction of CD86 is associated with diseases such as gallbladder squamous cell carcinoma and myocarditis.
