Target Information
The PAX genes are a family of transcription factors active in specific tissues during early embryonic development. These genes contain a paired box domain responsible for DNA binding and transcriptional regulation.
Several PAX genes are linked to developmental disorders:
- PAX3: Associated with Waardenburg syndrome
- PAX6: Associated with Aniridia
- PAX2: Associated with Wilms tumor
During embryogenesis, PAX2 is expressed in the developing kidney, particularly in the condensing metanephric mesenchyme and early epithelial structures derived from mesenchyme. Expression is downregulated as the tubular epithelium matures. However, PAX2 expression persists in undifferentiated epithelium of Wilms tumors, correlating with proliferation of poorly differentiated epithelial cells.
Interestingly, expression of the Wilms tumor suppressor protein WT1 coincides with downregulation of PAX2, suggesting that WT1 may directly repress PAX2 transcription.
