Target Information
STAT3는 STAT (signal transducers and activators of transcription) 단백질군에 속하며, 수용체 관련 키나아제에 의해 인산화되어 핵으로 이동하고, 사이토카인 및 성장인자에 반응하여 전사 인자로 작용합니다. CREB-binding protein 등의 공동활성화 인자가 STAT3의 전사 활성화에 필요합니다.
STAT3는 Interferon-α, Interferon-γ, EGF, PDGF, IL6 등에 의해 활성화될 수 있으며, Tyr705 위치의 인산화는 JAK1 및 JAK2에 의해 매개되어 이합체 형성, 핵 이동, DNA 결합에 필수적입니다.
STAT3 유전자는 인간 17번 염색체 q팔에 위치하며, 세포 성장과 세포사멸 등 다양한 세포 과정에 핵심적인 역할을 합니다. Rac1은 STAT3의 활성을 조절하며, PIAS3는 STAT3의 특이적 억제 단백질입니다.
STAT3는 세 가지 대체 스플라이싱 전사체를 가지며, knockout 마우스에서는 배아 초기 단계에서 치명적인 결손을 보입니다.
