Target Information

MLCK (also known as MYLK3) is a member of the myosin regulatory light chain kinases that facilitate muscle contraction. It belongs to the Ser/Thr protein kinase family and is responsible for smooth muscle contraction through phosphorylation of a specific serine in the N-terminus of myosin light chains (MLC), promoting interaction with actin filaments.
MLCK plays a key role in vascular permeability and tissue edema formation. In the nervous system, it regulates astrocytic process growth and neurotransmitter release at sympathetic synapses. It is also involved in fibroblast apoptosis signaling.
Both smooth muscle and non-muscle isoforms are widely expressed in adult and fetal tissues, with non-muscle isoform 2 being predominant. The Telokin isoform, which binds calmodulin, is also broadly expressed. MLCK likely undergoes down-regulation via phosphorylation and contains one fibronectin type III domain and nine immunoglobulin-like C2-type domains.
