Target Information

Transforming Growth Factor (TGF) betas mediate many cell-to-cell interactions that occur during embryonic development. Three TGF betas (TGF beta 1, TGF beta 2, and TGF beta 3) have been identified in mammals. Each is synthesized as a precursor protein that is cleaved to yield a 112 amino acid polypeptide associated with the latent portion of the molecule.
TGF beta polypeptides are multifunctional, influencing cell proliferation, differentiation, and other functions across a wide range of cell types. Both transformed and nonneoplastic tissues release transforming growth factors, and nearly all mammalian cells possess specific TGF receptors.
The multimodal nature of TGF beta is reflected in its ability to stimulate or inhibit cellular proliferation—mesenchymal cells are generally stimulated, while epithelial or neuroectodermal cells are inhibited.
TGF beta 1, TGF beta 2, and TGF beta 1.2 exhibit similar biological activities, though receptor binding affinities may vary. TGF beta 2 is produced by many cell types and is found in high concentrations in porcine platelets and mammalian bone. Latent TGF beta 2 is the predominant isoform in body fluids such as amniotic fluid, breast milk, and the aqueous and vitreous humor of the eye.
