Product Specific Information
On Western blotting, this antibody detects a protein with an apparent molecular weight of 165 kDa.
Positive controls: Insulin (50 nM for 5 min) or TPA (100 ng/mL for 30 min) treated human embryonic kidney cells (293T) or CHO-T cells transiently transfected with a plasmid encoding human IRS1.
Target Information
IRS-1 is a major substrate of the insulin receptor and is phosphorylated in response to insulin, IGF-1, and IL-4 stimulation.
It undergoes phosphorylation on serine, threonine, and tyrosine residues in various tissues.
Casein kinase II mediates insulin-stimulated serine/threonine phosphorylation of IRS-1, with Thr 502 and Ser 99 identified as major phosphorylation sites.
IRS-1 contains three putative 14-3-3 binding sites (Ser 270, Ser 374, Ser 641), with the Ser 270 motif located in the phosphotyrosine binding domain responsible for insulin receptor interaction.
