Target Information
The Lipin family of nuclear proteins includes Lipin-1, Lipin-2, and Lipin-3, which contain a nuclear signal sequence, a conserved amino-terminal (NLIP) domain, and a carboxy-terminal (CLIP) domain.
LPIN1 (Lipin-1) is essential for normal adipose tissue development and metabolism. It activates PGC-1A target pathways such as fatty acid oxidation and mitochondrial oxidative phosphorylation by inducing PPARA expression. LPIN1 also suppresses lipogenic programs and circulating lipid levels.
Deficiency in LPIN1 can lead to impaired adipose tissue development, insulin resistance, and lipodystrophy, characterized by loss of body fat, fatty liver, hypertriglyceridemia, and insulin resistance.
