Target Information
CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4)은 130 kDa 막당단백질로, scavenger receptor cysteine-rich (SRCR) 초가족에 속하며 hemoglobin-haptoglobin 복합체의 수용체로 작용합니다.
이 단백질은 자유 헤모글로빈으로 인한 산화 손상으로부터 조직을 보호하며, 헤모글로빈/하프토글로빈의 엔도사이토시스를 통해 철의 흡수 및 재활용에 관여합니다.
CD163은 인간 단핵구 및 대식세포 표면에 특이적으로 발현되며, 염증 후기 단계에서 주로 나타납니다. CD14 양성 단핵구, 대부분의 CD64 양성 단핵구, 그리고 CD16 양성 단핵구에서 높은 발현을 보입니다.
IL-10, IL-6, dexamethasone에 의해 발현이 증가하며, LPS 및 PMA 처리 시 세포 표면에서 shedding되어 혈장 또는 세포 상등액으로 방출됩니다.
CD163은 칼슘 및 pH 의존적으로 hemoglobin/haptoglobin 복합체를 결합하며, HP1F 형의 다중체 하프토글로빈 복합체에 대해 더 높은 친화성을 가집니다. 또한, tyrosine kinase 의존적 신호전달 경로를 통해 칼슘 이동, inositol triphosphate 생성, IL-6 및 CSF1 분비를 유도합니다.
