Target Information
PUMA는 Bax 및 Noxa를 포함한 pro-apoptotic Bcl-2 family의 구성원으로, p53의 전사적 표적입니다. PUMA 유전자는 BH3 도메인을 포함하는 두 단백질(PUMA-a 및 PUMA-b)을 암호화합니다. 이 단백질들은 Bcl-2에 결합하고, 미토콘드리아에 위치하며, p53에 의해 유도된 cytochrome c 방출 및 apoptosis를 유발합니다. PUMA는 p53 유도성 apoptosis의 직접적인 매개체로 작용할 수 있으며, PUMA 발현 억제 시 p53에 대한 세포의 apoptotic 반응이 감소하는 것이 관찰되었습니다. 따라서 PUMA는 cytochrome c/Apaf-1 의존적 경로를 통한 p53 유도 세포사멸에 중요한 역할을 하는 것으로 알려져 있습니다.
