• R932-25 was previously sold under product #46-0284. Use #R932-25 for re-ordering.
• Detects 6x His tag (C-term) -His-His-His-His-His-His-COOH; requires free carboxyl group for detection (Lindner et al., 1997).
• Contains enough material for 25 Western blots.
• Western blot with 100 ng recombinant Positope™ control protein gives detectable signal within 15 minutes using chromogenic substrate.
• Positope™ control protein (53 kDa) contains seven epitope tags, including His(C-term), HisG, c-myc, and V5.
• Important: Do not dilute AP-conjugated antibody in PBS (phosphate inhibits alkaline phosphatase).
  • Use Tris-Buffered Saline (TBS) with 0.1% Tween-20 and 1% nonfat dry milk.
  • Other blocking agents such as BSA or gelatin may be used.
• All AP-conjugated antibodies tested in immunoblotting with low background and no cross-reactivity in bacterial lysates.

Target Information

The 6x His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH).
Commonly expressed at N- or C-terminus of recombinant proteins for purification via immobilized metal affinity chromatography.
Epitope tagging enables detection of recombinant proteins without specific antibodies.
His-tagged proteins are purified by metal chelate chromatography, typically using Ni²⁺-NTA adsorbent due to strong and selective histidine–metal binding.
His tag-specific antibodies facilitate detection and co-immunoprecipitation of tagged proteins.
