Product Specific Information

This 1RNM44N monoclonal antibody recognizes mouse CD170 (Siglec F), a cell surface lectin of the Ig superfamily that binds sialic acid-containing glycoconjugates. Siglec F is mainly expressed on eosinophils and alveolar macrophages, with lower levels on immature myelomonocytic cells. It serves as a functional ortholog of human Siglec 8 but is not expressed on mast cells.

Each product includes:

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse thioglycolate-elicited peritoneal exudate cells. Recommended ≤0.1 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize titration for best performance.

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

Blocking Buffer Usage:
CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining. Add 5 µL per sample before combining with cells. For single-color controls, use 5 µL per 100 µL cell sample.

Target Information

SIGLEC5 is a putative adhesion molecule mediating sialic acid-dependent binding to cells. It binds both α2,3- and α2,6-linked sialic acid. Expression is found in monocytes and neutrophils, with high levels in peripheral blood leukocytes, spleen, and bone marrow, and lower levels in lymph node, lung, and other tissues.
