Product Specific Information

This 1RNM44N monoclonal antibody recognizes mouse CD170 (Siglec F), a cell surface lectin belonging to the Ig superfamily. Siglec F binds glycoconjugates containing sialic acids and is mainly expressed on eosinophils and alveolar macrophages. Lower expression is also observed on immature myelomonocytic cells. Mouse Siglec F acts as the functional ortholog of human Siglec 8 but is not expressed on mast cells.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse thioglycolate-elicited peritoneal exudate cells at ≤0.1 µg/test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Antibody titration recommended for optimal performance.

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining. Add 5 µL per sample or per 100 µL cell suspension (10³–10⁸ cells).

Target Information

SIGLEC5 is a putative adhesion molecule mediating sialic acid-dependent binding. It binds both α2,3- and α2,6-linked sialic acids. Expression is high in peripheral blood leukocytes, spleen, and bone marrow, and lower in lymph node, lung, appendix, placenta, pancreas, and thymus. It is expressed by monocytes and neutrophils but absent in early leukemic myelomonocytic cell lines.
