Target Information

The 26S proteasome is a multicatalytic proteinase complex with a highly ordered structure composed of two complexes: a 20S core and a 19S regulator.

  • The 20S core consists of four rings of 28 non-identical subunits (two rings of seven alpha subunits and two rings of seven beta subunits).
  • The 19S regulator is composed of a base (six ATPase subunits and two non-ATPase subunits) and a lid (up to ten non-ATPase subunits).
    Proteasomes are abundant in eukaryotic cells and cleave peptides in an ATP/ubiquitin-dependent, non-lysosomal pathway.
    A modified form, the immunoproteasome, processes class I MHC peptides.
    The PSMD2 gene encodes one of the non-ATPase subunits of the 19S regulator lid and may also participate in the TNF signaling pathway by interacting with the TNF type 1 receptor.
    A pseudogene has been identified on chromosome 1.
