Target Information

The 26S proteasome is a multicatalytic proteinase complex composed of a 20S core and a 19S regulator.

  • The 20S core consists of 4 rings of 28 non-identical subunits (2 rings of 7 alpha and 2 rings of 7 beta subunits).
  • The 19S regulator includes a base (6 ATPase and 2 non-ATPase subunits) and a lid (up to 10 non-ATPase subunits).
    Proteasomes are abundant in eukaryotic cells, cleaving peptides in an ATP/ubiquitin-dependent process.
    The immunoproteasome variant processes class I MHC peptides.
    PSMD2 encodes a non-ATPase subunit of the 19S regulator lid and may also play a role in TNF signaling through interaction with TNF receptor type 1.
    A related pseudogene exists on chromosome 1.
