Product Specific Information

The monoclonal antibody 20Raj1 recognizes the human and canine Ki-67 protein.
Ki-67 exists in two isoforms (345 and 395 kDa) expressed in dividing cells and is detectable during active phases of the cell cycle (G1, S, G2, M) but absent in resting (G0) cells.
During interphase, Ki-67 localizes to the nucleus and redistributes to chromosomes during mitosis, associating with heterochromatin-bound proteins like CBX3.
In tumor studies, Ki-67 expression serves as a proliferation marker indicating the fraction of dividing cells.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-diluted and validated using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
Recommended use: 5 µL (0.03 µg) per 100 µL test volume, with 10⁵–10⁸ cells/test (empirically determined).

Fluorochrome Information:
Brilliant Violet™ 480 emits at 479 nm and is suitable for cytometers with a violet (405 nm) laser.
Use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) when combining polymer dye-conjugated antibodies.
Not compatible with methanol-based fixation.

Target Information

Ki-67 is a nuclear protein expressed during G1, S, G2, and M phases of the cell cycle.
It associates with euchromatin and nucleolar structures through its FHA domain.
Expression is perinucleolar at G1, nuclear matrix-associated at G2, and chromosome-associated during mitosis.
Ki-67 is widely used as a proliferation marker in tumor studies, correlating with tumor grade and growth fraction.
It is also used as a neuronal marker of cell cycling and proliferation.
