Target Information
Post Synaptic Density 95 kDa (PSD-95), also known as synapse-associated protein 90 kDa (SAP90), is a membrane-associated protein found in the postsynaptic density of forebrain neurons and certain presynaptic structures in the cerebellum.
PSD-95 contains three PDZ domains, an SH3 domain, and a guanylate kinase homology (GuK) domain. It plays a role in clustering NMDA receptors and Shaker-type potassium channels at the synaptic membrane in CNS neurons.
PSD-95 interacts with other proteins through:
- C-terminal T/SXV motifs shared by NMDA receptors, β1 adrenergic receptor, some serotonin receptors, and sodium/potassium channel subunits.
- PDZ domain interactions, such as with neuronal nitric oxide synthase (nNOS), which associates via a pseudo-homotypic PDZ–PDZ interaction.
