Product Specific Information

The CD28.2 monoclonal antibody reacts with the human CD28 molecule, a 44 kDa homodimer expressed by thymocytes, mature T cells, and plasma cells. CD28 acts as a ligand for CD80 (B7-1) and CD86 (B7-2), providing potent co-stimulation of T cells. Signaling through CD28 enhances IL-2 and IL-2 receptor expression and cytotoxicity of CD3-activated T cells.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Use 5 µL (0.8 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology, utilizing nucleic acid dye structures engineered for optimized fluorescent signatures with reduced spillover and spread.

Target Information

CD28 antigen is a 44 kDa disulfide-linked homodimeric T-cell surface glycoprotein belonging to the immunoglobulin superfamily. It is constitutively expressed on most peripheral blood T lymphocytes and provides a critical co-stimulatory signal by binding CD80 and CD86 on antigen-presenting cells. CD28 prevents T cells from entering an anergic state or premature apoptosis. In murine lymphoid tissues, CD28 is expressed on all CD4+ and CD8+ T cells, with highest expression on immature thymocytes.
Diseases associated with CD28 dysfunction include mycosis fungoides and Sezary’s disease.
