Product Specific Information

The CD28.2 monoclonal antibody reacts with the human CD28 molecule, a 44 kDa homodimer expressed by thymocytes, mature T cells, and plasma cells. CD28 acts as a ligand for CD80 (B7-1) and CD86 (B7-2), serving as a potent co-stimulator of T cells. Signaling through CD28 enhances IL-2 and IL-2 receptor expression and increases cytotoxicity of CD3-activated T cells.

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of normal human peripheral blood cells at 5 µL (0.8 µg) per test. A test corresponds to the amount of antibody that stains a cell sample in 100 µL volume. Cell count may range from 10⁵ to 10⁸ cells per test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining and should be added to antibody cocktails before combining with cells.
  • Add 5 µL per sample (regardless of the number of NovaFluors).
  • For single-color controls: 5 µL per 100 µL of cell sample containing 10³–10⁸ cells.

Target Information

CD28 antigen is a 44 kDa disulfide-linked homodimeric T cell surface glycoprotein of the immunoglobulin superfamily. It is constitutively expressed on most peripheral blood T lymphocytes and acts as a critical co-stimulatory receptor by binding CD80 and CD86 on antigen-presenting cells. CD28 prevents T cells from entering an anergic state or undergoing premature apoptosis. In murine lymphoid organs and blood, all CD4⁺ and CD8⁺ T cells express CD28. Expression is highest on immature thymocytes and decreases on mature cells. CD28 dysfunction is associated with diseases such as mycosis fungoides and Sezary’s disease.
