Target Information

The NF-kB/Rel transcription factors are present in the cytosol in an inactive state, complexed with inhibitory IkB proteins. Activation of NF-kB occurs through phosphorylation-induced, proteasome-mediated degradation of IkB. This process involves activation of the IKK complex, composed of IKK alpha, IKK beta (catalytic subunits), and IKK gamma (regulatory subunit). Phosphorylation at Ser177 and Ser181 in IKK beta (Ser176 and Ser180 in IKK alpha) triggers conformational changes leading to kinase activation.
