TCTN3는 Tectonic 단백질 패밀리에 속하며, Hedgehog (Hh) 신호전달 경로를 통한 신경관 패터닝 조절에 관여하는 진화적으로 보존된 분비형 및 막관통 단백질입니다.
TCTN3의 발현 패턴과 기능은 명확히 밝혀지지 않았으나, TCTN1과 58%의 유사성을 보입니다.
TCTN1은 배아 발생 동안 Hh 신호전달에 관여하는 부위에서 발현되며, TCTN1 결손 마우스는 E13.5~E16.5 사이에 사망하고 holoprosencephaly를 나타냅니다. 이는 TCTN1이 Hh 활성화 인자로 작용함을 시사합니다. 후기 발생 단계에서는 전후 신경관에서 Hh 경로의 억제 인자로 작용할 가능성이 있습니다.
