SMADs are members of the MAD-related family of intracellular proteins that play essential roles in the signaling pathways of serine/threonine kinase receptors of the TGF-β superfamily.
They are categorized into:
- R-SMADs: SMAD1, SMAD2, SMAD5, SMAD8, SMAD9
- Co-SMAD: SMAD4
- I-SMADs: SMAD6, SMAD7
SMAD1, SMAD5, SMAD8, and SMAD9 share high sequence homology, and antibodies recognizing common sequences are available. SMAD8 and SMAD9 are often used interchangeably in literature.
