Target Information
KLOTHO is a systemic anti-aging hormone within the glycosidase1 superfamily. It encodes a type I membrane protein that is abundant in the kidney and brain. In mice, deficiency in KLOTHO expression leads to aging-like phenotypes such as arteriosclerosis, osteoporosis, and skin atrophy, along with growth retardation, short lifespan, and infertility. Transgenic mice overexpressing KLOTHO show an extended lifespan by inhibiting insulin/IGF1 signaling. KLOTHO is also involved in regulating calcium and phosphorus homeostasis by inhibiting the synthesis of active vitamin D and has been identified as a potential tumor suppressor.
