Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events related to pregnancy establishment and maintenance.
PR exists as two forms: a 94 kDa protein (Form A) and a 120 kDa protein (Form B), generated by alternative translation initiation sites.

  • PR-B: Transcriptionally active form responsible for activating genes for endometrial maintenance, pregnancy, and inhibition of ovulation.
  • PR-A: Lacks 165 amino acids at the N-terminus, exposing a 140 amino acid inhibitory domain that represses steroid hormone transcriptional activity.

In its inactive state, PR forms a multiprotein complex including heat shock proteins and immunophilins. Upon progesterone binding, the receptor undergoes conformational change, dimerizes, and binds to progesterone response elements (PRE), leading to activated transcription.
A null mutation in the PGR gene results in pleiotropic reproductive abnormalities.
