Target Information

Phosphoinositide-specific phospholipase C (PLC) plays a crucial role in receptor-mediated signal transduction by generating two second messengers, inositol 1,4,5-triphosphate and diacylglycerol, from phosphatidylinositol 4,5-bisphosphate. Mammalian PLC isozymes include PLCβ1–4, PLCγ1–2, PLCδ1–2, and PLCε.

PLCδ1 acts as a calcium signal amplifier and is activated by an atypical GTP-binding protein. It is an effector for transglutaminase II-mediated oxytocin receptor and α1B-adrenoreceptor signaling. Mouse PLCδ1 is highly expressed in brain, heart, lung, and testis. Abnormal accumulation of PLCδ in Alzheimer’s disease (AD) brains suggests a role in AD pathology.

PLCδ2 is markedly expressed in type II intestinal metaplasia and adenocarcinoma. Its presence in type I intestinal metaplasia is associated with benign states but may evolve toward neoplastic transformation, indicating PLCδ2 expression could serve as a marker of gastric malignant transformation.
