Target Information
Phosphoinositide-specific phospholipase C (PLC)는 receptor-mediated signal transduction의 시작에서 중요한 역할을 하며, phosphatidylinositol 4,5-bisphosphate로부터 inositol 1,4,5-triphosphate와 diacylglycerol을 생성하여 두 개의 세컨드 메신저를 형성합니다.
PLC에는 여러 동형이 존재하며, PLC beta1, beta2, beta3, beta4, gamma1, gamma2, delta1, delta2, 그리고 PLCε가 포함됩니다.
PLC delta는 네 가지 isoform으로 존재하며, PLC delta1은 calcium signal amplifier로 작용하고, GTP-binding protein transglutaminase II를 통한 oxytocin receptor 및 alpha1B-adrenoreceptor 신호전달의 effector 역할을 합니다.
PLC delta1은 뇌, 심장, 폐, 고환에서 높은 발현을 보이며, Alzheimer’s disease(AD) 환자의 뇌에서 비정상적으로 축적되어 AD 병리와 관련이 있을 가능성이 있습니다.
PLC delta2는 type II intestinal metaplasia와 adenocarcinoma에서 현저히 발현되며, type I intestinal metaplasia에서 발현될 경우 양성으로 간주되지만 종양성 변환으로 진행될 수 있습니다. 따라서 PLC delta2 발현은 위암성 변형의 잠재적 표지자로 고려될 수 있습니다.
