Target Information
MERTK (cMER) is a tyrosine kinase proto-oncogene involved in the retinal pigment epithelium (RPE) phagocytosis pathway, which is implicated in human retinal disease. It regulates physiological processes including cell survival, migration, differentiation, and efferocytosis (phagocytosis of apoptotic cells). Ligand binding induces autophosphorylation of MERTK, creating docking sites for downstream signaling molecules such as GRB2 or PLCG2, leading to phosphorylation of MAPK1, MAPK2, FAK/PTK2, or RAC1. MERTK signaling contributes to macrophage clearance of apoptotic cells, platelet aggregation, cytoskeleton reorganization, and inhibition of TLR-mediated innate immune responses via STAT1 activation and SOCS1/3 induction.
