Perforin은 표적 세포의 삼투 용해를 유도하는 pore-forming protein으로, granzymes가 세포 내로 침투하여 apoptosis를 유도하도록 합니다. C9과 구조적·기능적 유사성을 가지며, 다양한 표적 세포를 용해할 수 있습니다. T세포 및 NK세포 매개 세포용해에 핵심적인 효과 분자이며, PRF1 유전자 결함은 어린 시절 발병하는 치명적 유전 질환인 가족성 혈구탐식성 림프조직구증(type 2)을 유발합니다. 활성화된 CD8+, NK 및 일부 CD4+ T세포에서 perforin 발현이 증가합니다.
Target Information
Perforin은 세포독성 과립의 주요 단백질로, 세포독성 림프구에 의해 종양 및 바이러스 감염에 대한 면역 방어에 관여합니다.
- 단백질 길이: 555 amino acids (signal peptide 21 aa 포함)
- 분자량: 70–75 kDa
- C9과 유사한 transmembrane channel 형성 메커니즘을 가짐
- Killer cell line에서만 발현, helper T lymphocytes 및 기타 종양세포에서는 발현되지 않음
- PRF1 유전자 결함은 가족성 혈구탐식성 림프조직구증(type 2)을 초래
- Alternative splicing으로 다양한 전사체 존재
