Product Specific Information

  • Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

Calcineurin is a Ser/Thr, calcium and calmodulin-dependent protein phosphatase essential in the T cell activation pathway. It consists of two subunits:

  • Calcineurin A (CnA): 60 kDa catalytic subunit, homologous to protein phosphatases 1 and 2A.
  • Calcineurin B (CnB): 19 kDa regulatory subunit containing four EF-hand calcium binding sites.

Calcineurin-mediated dephosphorylation of NF-AT is crucial for its activation and nuclear translocation. Immunosuppressive drugs such as cyclosporin A and FK506 inhibit this process by binding to immunophilins (cyclophilin and FKBP), forming complexes that block substrate access to the catalytic domain of CnA without inhibiting its phosphatase activity.
