The Human MERS S1 Ig Total Simplex ProcartaPlex Kit measures anti-MERS S1 antibodies in serum and plasma. The detector antibody is an Immunoglobulin (Ig) Total antibody directly conjugated to phycoerythrin (PE) that cross-reacts with all Ig (IgG, IgM, and IgA) in serum and plasma. IgG is the most abundant of the immunoglobulins.

This kit can be combined with other Simplex kits from the Coronavirus Panel to create a custom multiplex panel utilizing Luminex xMAP technology for protein detection and quantitation. When combining multiple Simplex kits, only one Viral Reagent kit (sold separately) is required per assay plate regardless of plex size.

About ProcartaPlex Assays for the Luminex Platform

ProcartaPlex immunoassays are based on the sandwich ELISA principle, using two highly specific antibodies binding to different epitopes of one protein to quantify multiple targets simultaneously using a Luminex instrument.

ProcartaPlex assays utilize Luminex xMAP (multianalyte profiling) technology for simultaneous detection and quantitation of up to 80 protein targets in a single 25–50 μL sample from plasma, serum, cell culture supernatants, and other bodily fluids.

The Luminex MagPlex superparamagnetic microsphere beads in the ProcartaPlex assay are internally dyed with precise proportions of red and infrared fluorophores to create 100 spectrally unique bead signatures identifiable by Luminex xMAP detection systems (Luminex 200, FLEXMAP 3D, and MAGPIX). Similar to a sandwich ELISA, the ProcartaPlex assay uses matched antibody pairs to identify the target protein. Each bead, labeled with a specific antibody, binds its target protein, which is then detected using biotinylated antibodies and streptavidin–R-phycoerythrin (RPE).

The key difference between a ProcartaPlex assay and ELISA is that the capture antibody in ProcartaPlex is conjugated to a magnetic bead rather than adsorbed to a microplate well, allowing reagents to remain free-floating. The Luminex instrument uses two lasers—one to identify bead spectral signatures and another to quantify RPE fluorescence, proportional to protein concentration. ProcartaPlex assays profile more targets with less sample in the same time as a traditional ELISA.

ProcartaPlex multiplex panels are available for multiple species (human, mouse, rat, nonhuman primate, porcine, and canine).
