TLE1은 다양한 전사 인자에 결합하는 전사 억제 단백질입니다. NF-kappa-B에 의해 조절되는 유전자 발현을 억제하며, FOXA2, CTNNB1, TCF family에 의한 전사 활성화를 억제합니다. AES와의 상호작용에 의해 TLE family의 기능이 조절될 수 있으며, ESRRG의 공동활성인자로 작용하는 특이한 기능을 가집니다.
