Target Information
LCK is a member of the Src-family tyrosine kinases essential for signaling through the T-cell receptor (TCR) complex. Upon TCR activation, LCK phosphorylates ITAM motifs in zeta subunits, enabling ZAP70 binding and activation. Activated ZAP70 phosphorylates adaptor LAT to propagate downstream signaling.
Most LCK localizes to the plasma membrane, with a portion associated with the Golgi apparatus contributing to Raf activation during weak TCR stimulation. LCK has N-terminal myristylation and palmitylation sites, a PTK domain, and SH2/SH3 domains mediating protein-protein interactions.
LCK is also involved in apoptosis regulation independent of death receptor pathways. Diseases associated with LCK include Immunodeficiency 22 and CD45 deficiency. Alternative splice variants encoding different isoforms have been identified.
