Target Information
CD20 is a non-glycosylated surface phosphoprotein with a molecular weight range of 33–37 kDa depending on phosphorylation. It is expressed on mature and most malignant B cells, a subpopulation of T lymphocytes, and follicular dendritic cells. CD20 expression coincides with surface IgM and regulates calcium conductance, cell cycle progression, and B-cell proliferation.
CD20 associates with lipid rafts, and its interaction depends on extracellular triggering and BCR aggregation. Following receptor ligation, CD20 colocalizes with BCR but remains at the cell surface after BCR endocytosis. As CD20 is stably expressed on B-cell malignancies and not internalized after antibody treatment, it serves as a key target for therapeutic B-cell depletion.
Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
