Target Information
The CDKN2A gene produces several transcript variants differing in their first exons. At least three alternatively spliced variants encode distinct proteins, two of which function as inhibitors of CDK4 kinase. Another transcript encodes an alternate open reading frame (ARF) that produces a structurally unrelated protein functioning as a stabilizer of the tumor suppressor protein p53 by interacting with and sequestering MDM2. Despite structural and functional differences, both CDK inhibitor isoforms and the ARF product contribute to cell cycle G1 control through regulation of CDK4 and p53. This gene is frequently mutated or deleted in various tumors and is recognized as an important tumor suppressor gene.
